The injury forced him to miss the Barcelona tour of the United States (US) as well as the shocking defeat to Athletic Bilbao in their league opener nine days ago.
Messi had returned to training earlier this week only for him to feel discomfort in training on Friday thereby ruling him out of the Real Betis game.
Ernesto Valverde and his Barcelona players must be wary of the threat carried by Real Betis who were the only team to win at the Nou Camp last season.
Barcelona were beaten 4-3 by Real Betis in the corresponding fixture last season.
